    5779: What techniques can we use to detect amorphous/amorphous phase separation within ASDs (amorphous solid dispersions) where the active drug substance is not crystallising but the two amorphous components are phase separating?


    5776: We are increasingly using amorphous solid dispersions to formulate poorly soluble drug substances. How can we confirm that we have a fully homogenous molecular dispersion? Currently we use Raman imaging to confirm homogeneity but this is limited to approx 1um spatial resolution. The presence of phase separation at the sub-micron scale could potentially lead to instability and crystallisation of the drug substance.

    5779: Many of the techniques we use to confirm the homogeneity of amorphous solid dispersions are based on detection of residual crystalline drug e.g. cross-polar microscopy. How do we detect phase separation occurring when both components remain in the amorphous form.

    How would you visualise the assessment of microbial impact of a topical product on skin?


    Reckitt owns multiple brands that have on skin applications such as Dettol, Nurofen and Durex. Society is becoming increasing aware of the importance of maintaining healthy skin to protect themselves from external factors. Society is also becoming increasing aware of the components associated with healthy skin, such as a good strong physical barrier and a balanced microflora. Therefore, people are becoming more selective with the types of products they buy, which are applied directly to the skin as they don’t want to cause harm to their body. Not everyone’s microbiome is same and different microbes reside in different locations across the body. Existing methodologies to analyse the microbiome on the skin include taking swap samples however, this does not capture visual context of what is happening on the skin. Therefore, we would like to discuss how this is possible and level of detail we will be able to see.

    As in silico and in vitro models evolve, how close are we to correlating biological responses from these models to in vivo biological responses?


    Clinical trials are expensive and time consuming for companies. A clinical trial failure, where a trial fails to meet the success criteria, can set the company back financially and significantly delay product launches. Therefore, to increase the chances of clinical success, companies are using pre-clinical models to reduce the risk of failure i.e. in silico and in vitro models to predict desired biological responses. Until recently, however, these models have not always mimicked in vivo biological responses and therefore, have not reduced the risk of clinical trial failure. Hence, we would like to know with the advancement of model development, are these models able to better predict in vivo biological responses.
